I couldn't decide who I'd personally want, so I just put all 3 of them here.
These are the main females from the Mother series, if you were wondering.
Paula: Ness's friend girl (girlfriend) from Earthbound, able to use very helpful and deadly PSI attacks. Ness had to save her from Happy Happy Village, who had kidnapped her (and I don't remember why, even though I recently saved her TWICE since I'm playing through the game in two save files). After that, she gets kidnapped by Geralde Monotoli once the group goes to Fourside. Ness then saves her again, for the last time, thankfully. Paula needs to pray to defeat Giygas, making her extremely important to keep alive in that fight. She lives in a town called Twoson, directly south from Onett, Ness's hometown. She is famous for her psychic powers in Twoson, and apparently the "media" shows up at the Polestar Preschool to see her occasionally.
I'm suggesting Paula because Earthbound has a huge fanbase (OMG Starmen.net is too much), and Paula is very important to the game. In uniqueness, Paula probably wouldn't score that many points, though. I know about a million softhearted slightly dramatic girls in RPGs, and I know you do too. Paula's combat is unique compared to other RPG girls, because there are hardly any RPG girls that don't heal other teammates, but would rather severely kick their opponents' asses. I think her moves could look like they did in-game, like Lucas's Crusade-only moves (For Final Smash, think Pray. It only makes sense).

Ana: A girl in Mother/Earthbound Zero who knew that Ninten would show up at her door one day. She is incredibly sweet; she once saved frogs from being dissected at her school. Sadly, her mother has been abducted, with the same fate descending upon every child in the town of Youngtown. She definitely does not disappoint; she can learn tons and tons of PSI spells, and all you have to do is hardly level her up a few times. She's great and the only downside is her tendency to be a target for enemies (FREAKING EAGLES IDOJAMISXLJIWI). Ana openly expresses her love for Ninten when she asks him to dance with her. Also, Ana is Catholic and reads the Bible often, making her better than any other dumb non-Christian video game characters.
Ana is a lot like Paula, with few differences. The PSI she learns is the biggest difference. Ana can learn a crapload of PSI, from PK Beam to Brainshock to PK Thunder to PSI Block to blah blah blah blah... Therefore, she has the same crapload of variety possible in her moveset (For Final Smash, think PK Beam Gamma. In Mother, if it hit, it would IMMIDIATELY defeat any enemy, except maybe bosses of course). For originality, I won't give much other than the Bible part. I've NEVER seen a Christian video game character but Ana.
Kumatora: The spunky and tomboyish princess of Osohe Castle. She was first seen when the player was controlling Duster, on a theif mission. Kumatora dropped a pendant as she was running, which Duster picked up and brought back to his father, Wess. Immediately recognizing the pendant, both Wess and Duster returned to Osohe to find Kumatora with her leg caught in a "busted-ass trap." After travelling in the castle for a while and finding the egg of light, Kumatora and Wess get separated from Duster. Later in the game, Kumatora works in a place called club Tittiboo as a waitress under the name Violet.
Kumatora is probably the most unique of all of these three. She's kind of the opposite of both Paula and Ana, cursing infrequently, getting impatient, not showing love for Lucas at all, and, basically, being very tomboyish. Her PSI techniques are quite similar to Paula's with a few differences (PK Ground would have been something unique if Lucas didn't steal it. AND Starstorm. FFFFFFFFFFFF-. At least she knows Lifeup. =p). Plus, she scores many point for uniqueness. I can't think of another girl in an RPG that joins your group like this one. She even uses different weapons than Paula and Ana, who use frying pans whereas Kumatora uses gloves. Overall, she's very cool, and "cool" is not something I'd call Ana or Paula. I'd call them sweet, nice, or awesome, but Kumatora gets cool.
